					<p>Alle in dieser Arbeit verwendeten Peptide wurden hergestellt durch Dr. Achim Kramer, Institut für Medizinische Immunologie, Universitätsklinikum Charité, Humboldt-Universität, D-10117 Berlin.<br/>Als Ausgangspeptid und Positivkontrolle wurde das Peptidepitop (323-339) des Ovalbumin-Proteins aus Hühnereiweiss (kurz: OVA<sub>323-339</sub>) verwendet. Um die Spezifität der beobachteten Reaktionen zu überprüfen, wurde in den <em>in vitro</em> Experimenten Motten-Cytochrom-C (MCC) (88-103) als alternativer Antigenstimulus angeboten. Für die i.v. Injektionen wurden die Peptide in PBS (in der Regel 1mg Peptid/ml PBS) gelöst. Die Proben wurden bei -20°C gelagert.<br/>In der Tabelle unten sind die in den Experimenten verwendeten Peptide im IUPAC-Einbuchstaben-Code (IUPAC= engl. International Union of Pure and Applied Chemistry) dargestellt siehe auch <link ref="_Ref49172222">Tabelle 10.1</link>.</p>

					<p>Für das zunächst erfolgende Screening (Schnelltest) in der Verkürzungsanalyse ( engl. Truncationanalysis)und bei der Substitutionsanalyse wurden die Peptide auf einer Zellulosematrix synthetisiert (siehe auch Grogan und Kramer 1999 [<link ref="_bib78">81</link>]). Nach der Synthese wurden die Peptide <pagenumber id="N111EE" label="30" numbering="arabic" start="30"/>von der Zellulose durch Behandlung mit gasförmigen NH<sub>3</sub> abgespalten und die Spots ausgestanzt. Die ausgestanzten Spots wurden anschließend in sterilem Aqua bidest (1ml/Peptid/Spot) gelöst und archiviert. Die Peptid-Spots wurden bei -20°C verwahrt. <br/>Für die Titrationsversuche sowie für die <em>in vivo </em>Versuche wurden die ausgewählten Peptide konventionell synthetisiert, aufgereinigt und mittels HPLC und Massenspektrometrie charakterisiert.</p>
