         <p><citenumber helper="true" id="N13047" start="97"/>Die permanente, parathyreoprive Hypokalzämie stellte eine schwerwiegende Komplikation in der Schilddrüsenchirurgie dar. </p>
         <p>Die derzeitige Therapie der Wahl ist die lebenslange Substitution von Vitamin D und Kalzium. Aufgrund der komplexen Stoffwechselwirkungen des Parathormons zeigt diese jedoch teilweise unbefriedigende Ergebnisse.</p>
         <p>
            <citenumber id="N1304F" start="98"/>Bisherige Versuche der Allotransplantation von Parathyreozyten scheiterten letztlich immer an der Rejektion des Gewebes durch den Empfängerorganismus.</p>
         <p>Die Mikroverkapselung von Zellen mittels einer semipermeablen Membran bietet einen wirkungsvollen Schutz vor immunologischer Abstoßung bei freiem Substrataustausch und ermöglicht so die Transplantation ohne Immunsuppression.</p>
         <p>Bei den meisten der bisherigen Anwendungen zur Enkapsulierung von Zellen wird ein System aus Alginat / Polylysin verwendet. Aufgrund mitogener und toxischer Verunreinigungen der Alginate ist für die klinische Anwendung jedoch ein aufwendiges Reinigungsverfahren des Ausgangsproduktes notwendig.</p>
         <p>
            <citenumber id="N1305B" start="99"/>Mikrokapseln aus den Komponenten Natriumcellulosesulfat und Polydiallyldimethylammoniumchlorid (Poly-DADMAC) werden seit den neunziger Jahren erfolgreich auf vielen Gebieten zur Immobilisierung von Zellen verwendet. Bisherige Studien konnten keinerlei mitogene oder toxische Eigenschaften nachweisen. Die Enkapsulierung mit diesem Verfahren erfolgt über ein unkompliziertes, gut reproduzierbares Ein-Schritt-System.</p>
         <p>Ziel dieser Arbeit war es, die Eignung des Systems Natriumcellulosesulfat / Poly-DADMAC für die Mikroverkapselung von kryokonservierten, humanen Parathyreozyten zu prüfen.</p>
         <p>Insbesondere sollten hierbei die mechanischen Eigenschaften der hergestellten Mikrokapseln sowie die Viabilität und Funktion der enkapsulierten Zellen verifiziert werden.</p>
         <p>
            <citenumber id="N13067" start="100"/>Es konnte gezeigt werden, dass </p>
         <p>
            <ul>
               <li>
                  <p>die hergestellten Kapseln aus NaCS / Poly-DADMAC mit einer zum Zerplatzen aufzuwendenden Kraft F von 2,84 ± 1,36 N über eine hohe mechanische Festigkeit verfügen.</p>
               </li>
               <li>
                  <p>die Kapselmembran mit einer mittleren Trenngrenze (Maß für die Porengröße) von 1,39  0,04 nm den Erfordernissen einer semipermeablen und immunoprotektiven Barriere entspricht.</p>
               </li>
               <li>
                  <p>die Größe der hergestellten Kapseln relativ einheitlich ist (mittlerer Durchmesser 0,33  0,02 mm).</p>
               </li>
               <li>
                  <p>durch die Verkapselung von Parathyreozyten mit NaCS / Poly-DADMAC kein schädigender Einfluss auf die enkapsulierten Zellen festzustellen ist.</p>
               </li>
               <li>
                  <p>die Kalziumsensitivität der Parathyreozyten im Kalziumsuppressionstest auch nach Verkapselung der Zellen erhalten bleibt.</p>
               </li>
               <li>
                  <p>mit NaCS / Poly-DADMAC enkapsulierte Parathyreozyten im Vergleich zu unverkapselten Zellen ein Ansteigen der PTH-Werte in Zellkultur zeigen. </p>
               </li>

         <p>Es konnte somit in vitro die Eignung von Natriumcellulosesulfat und Poly-DADMAC zur Mikroverkapselung von kryokonserviertem, humanem Nebenschilddrüsengewebe aufgezeigt werden.</p>
         <p>
