			<p>In dieser Arbeit werden zwei Modelle für oxidativen Stress untersucht: </p>

						<p>Inkubation isolierter Typ-II-Pneumozyten mit 0,5mM H<sub>2</sub>O<sub>2</sub> für maximal 60min.</p>

			<p>Im Modell 1 der subletalen Hyperoxie sollen die <em>in vivo</em> auftretenden Veränderungen, im Modell 2 gezielt die Effekte auf Typ-II-Zellen erfasst werden. </p>
			<p>Für beide Modelle ist wichtig abzuklären, welchen Einfluss die Kulturbedingungen auf die Typ-II-Zellen haben: Da Typ-II-Pneumozyten üblicherweise unter Basalbedingungen in Raumluft und 37°C auf Plastikschalen in der Kultur gehalten werden, haben wir zunächst untersucht, inwiefern die unphysiologisch hohen Sauerstoffkonzentrationen der Raumluft einen Stressfaktor für die Zellen darstellen. Gemessen wurde die gesamte antioxidative Kapazität, die Aktivität der antioxidativen Enzyme Catalase und Superoxiddismutasen (SOD) sowie die Expression von Hitzeschockproteinen (HSP) im Verlauf der Zellkultur. </p>
			<p>Es wird angenommen, dass bei <em>in vivo</em> Hyperoxie zunächst das antioxidative Schutzschild des Surfactants durchschlagen werden muss, bevor es zu einer oxidativen Schädigung der Lunge kommt. Dabei stellt sich die Frage, ob die oxidative Schädigung der Lunge durch einen Mangel an funktionell wichtigen Lipiden und Proteinen oder durch zytotoxische Oxidations­produkte verursacht wird; weiterhin ist unklar, ob sich die oxidative Schädigung mehr im lipo- oder mehr im hydrophilen Milieu abspielt. Dies hat Auswirkungen auf die Ansätze einer möglichen klinischen Therapie. Dazu haben wir die Lipidperoxidation anhand der Konzen­tration von Lipidhydroperoxiden (LOOH), Malondialdehyd (MDA) und von PAF-ähnlichen Substanzen (PAF-RC) unter Hyperoxie und H<sub>2</sub>O<sub>2</sub>-Stress untersucht. </p>

				<pagenumber id="N1093A" label="22" numbering="arabic" start="22"/>Die PAF-Acetylhydrolase (PAF-AH) inaktiviert PAF-RC, die durch ROS im Rahmen der Entzündungsreaktion entstehen und proinflammatorische Wirkung haben (Frey 2000). Die ROS selbst jedoch können die PAF-AH-Aktivität rasch und irreversibel hemmen (Ambrosio 1994, Triggiani 1997). Da auch durch Entzündungsreaktionen ROS entstehen, wird durch den ROS-vermittelten Verlust der PAF-AH-Aktivität der proinflammatorische Effekt von PAF verlängert. Wir haben untersucht, inwieweit die Konzentration von PAF-RC in BAL und Plasma mit der Aktivität von PAF-AH korreliert und ob die ROS-induzierte Zerstörung von PAF-AH einen Mechanismus für Lungenschäden unter Hyperoxie oder H<sub>2</sub>O<sub>2</sub>-Behandlung darstellt. </p>
			<p>HSP sind in der Evolution stark konservierte Moleküle und werden in den meisten Zellen physiologisch auf niedrigem Niveau exprimiert. HSP sind wichtig für die Faltung von de novo synthetisierten Proteinen, deren Translokalisation durch intrazelluläre Membranen, für die Formation von Proteinoligomeren und deren Abbau. Bei oxidativer Belastung können sie beschädigte Proteine stabilisieren und neu falten. Hitzeschockproteine sind frühe und sehr sensitive Marker des zellulären Stress in vielen Zelltypen (Hightower 1991, Jaattela 1992). Ein Anstieg der HSP-Expression unter Hyperoxie ist bekannt (Choi 1995, Ho 1996), wir haben hier den Effekt von H<sub>2</sub>O<sub>2</sub> auf die Expression von HSP in Typ-II-Zellen untersucht. </p>
