         <head>Zielsetzung</head>
         <p>
            <citenumber id="N104E6" start="7"/>FGF&#8211;2 ist ein potenter proliferativer und mitogener Wachstumsfaktor. Er spielt eine Rolle bei der Pathogenese verschiedener Erkrankungen, unter anderem bei der Polypenbildung in der Nase, der Sklerodermie und der rheumatischer oder rheumatoider Arthritis. Dies sind Erkrankungen, die durch die pathologischen u.a. von Fibroblasten gekennzeichnet sind. Auch bei physiologischen Abläufen, wie beispielsweise der Wundheilung, ist FGF&#8211;2 von Bedeutung. MC sind vermehrt im Gewebe dieser Proliferationsprozesse vorhanden.<br/>Ziel dieser Arbeit ist die Untersuchung der FGF&#8211;2&#8211;Synthese von MC nach Stimulation mit verschiedenen Substanzen sowie Untersuchungen zur Sekretion von FGF&#8211;2 aus MC und der Sekretionsmodulation durch die Stimulantien und UV&#8211;Licht. MC wurden mit verschiedenen Substanzen inkubiert und anschließend die Menge des synthetisierten und freigesetzten FGF&#8211;2 ermittelt. Zu diesen Substanzen zählten die klassischen MC&#8211;Liberatoren (a&#8211;IgE, PMA und Ca&#8211;Ionophore), das MC aktivierenden Neurokinin Substanz P und einige proinflammatorische Zytokine (IL&#8211;4, IL&#8211;6, IL&#8211;8 und TGF&#946;). Für die Untersuchungen hinsichtlich des Einflusses von UV&#8211;Licht auf die FGF&#8211;2&#8211;Sekretion wurden die Zellen mit Licht spezifischer Wellenlängen bestrahlt und anschließend die Freisetzung ermittelt.<br/>Durch die hier gewonnenen Ergebnisse soll der Einfluss äußerer Faktoren auf MC und deren FGF&#8211;2 Synthese und Freisetzung gezeigt werden.</p>
