         <head>Zusammenfassung</head>
         <p><citenumber helper="true" id="N113AC" start="44"/>FGF&#8211;2 ist als Wachstumsfaktor an vielen verschieden Vorgängen des menschlichen Organismus beteiligt. Dazu gehören sowohl physiologische Prozesse wie die Wundheilung nach Verletzungen als auch vielfältige pathologische Prozesse, denen FGF&#8211;2 als pathogenetischer Faktor zugeordnet wird. Lange Zeit wurden Fibroblasten als Hauptproduzenten von FGF&#8211;2 angesehen, aber mittlerweile schließt die derzeitige Forschung auch MC als FGF&#8211;2&#8211;Produzenten ein. Damit gewinnen MC Einfluss auf die oben erwähnten physiologischen und pathologischen Prozesse.<br/>In dieser Arbeit wurden MC aus menschlichen Geweben isoliert und bezüglich der FGF&#8211;2&#8211;Synthese und &#8211;Freisetzung untersucht. Es wurde besonders die Syntheseleistung nach Stimulation mit verschiedenen Substanzen analysiert. Die Ergebnisse zeigten sowohl auf Protein&#8211;Ebene als auch auf mRNA&#8211;Ebene, dass die FGF&#8211;2&#8211;Synthese durch die verwendeten Substanzen moduliert wurde. Durch Stimulationen mit a&#8211;IgE, SP, IL&#8211;4, IL&#8211;6 und IL&#8211;8 wurde eine gesteigerte FGF&#8211;2&#8211;Synthese induziert. Weiterhin zeigten die vorliegenden Ergebnisse, dass bei der Degranulation der MC der Wachstumsfaktor freigesetzt wird. In einem weiteren Versuch wurde der Einfluss von UV&#8211;Licht auf die Freisetzung von FGF&#8211;2 untersucht. Hierbei zeigte besonders UVA<sub>1</sub>&#8211;und PUVA<sub>1</sub>&#8211;Bestrahlung einen inhibieren Effekt auf die Sekretion des Proteins. Zelleigene Proteasen scheinen keinen Einfluss auf den Wachstumsfaktor auszuüben.<br/>Die Ergebnisse weisen darauf hin, dass die FGF&#8211;2&#8211;Synthese in MC von äußeren Faktoren abhängig ist. MC können dadurch auf den Verlauf von pathologischen und physiologischen Vorgänge Einfluss nehmen. Die Funktion von MC im Immunsystem scheint sehr weitreichend und wirft weitere Fragen auf. Eine Modulation der FGF&#8211;2&#8211;Freisetzung durch UV Licht ergibt insbesondere bei Hauterkrankungen mögliche Aspekte die Wirkung der UV&#8211;Therapie weiter aufzuklären.</p>
