					<p>5-Fluorouracil (Roche Pharma, Reinau, CH) ist ein Pyrimidin Analog und wird im Körper über 5-FUMP zu 5-Fluorodeoxyuridinmonophosphat (FdUMP) umgewandelt, das eine hohe chemische Ähnlichkeit zu Thymidin aufweist. Als Antimetabolit wirkt FdUMP hemmend auf den DNA/RNA-Stoffwechsel. Es bindet die Thymidilat Synthase mit vielfach höherer Affinität als dUMP und blockiert damit den letzten Schritt der zellulären Thymidin Shynthese. Neben der DNA wird auch die RNA Synthese durch 5-FU gestört, indem zuerst gebildetes 5-FUMP in RNA eingebaut wird. Auch, wenn 5-Fluorouracil zu den alten Chemotherapeutika gehört, ist der klinische Einsatz insbesondere bei kolorektalen Tumoren auch heute noch aktuell.</p>
					<p>Paclitaxel (Bristol-Myer Squibb AG, Baar, CH) ist die als erste zugelassene Substanz der Taxane, einer neuen Zytostatika Klasse, die als Naturstoff aus der Rinde der pazifischen Eibe gewonnen wird. Durch Hemmung der Tubulindepolymerisierung wird die Organisation des Spindelapparats vor der Mitose verhindert. Der Zyklus der <pagenumber id="N10738" label="23" numbering="arabic" start="23"/>
betroffenen Zellen wird in der G2/M Phase blockiert. Zugelassen ist Taxol insbesondere für therpierefraktäre metastasierende Ovarial-/Mammakarzinom, verspricht allerdings auch bei anderen soliden Tumoren Erfolge (NSCLC, Kaposi Sarkom)</p>
				</subsection>
				<subsection id="N1073E" label="2.3.2">
					<head>Antisense Oligonukleotide</head>
					<p>4625: 5&#8217;-AAGGCATCCCAGCCTCCGTT-3&#8217; 4626: 5&#8217;-CACGTCACGCGCGCACTATT-3&#8217;</p>
					<p>Die Sequenz des 4625 Antisense Oligonukleotids richtet sich gegen eine Region hoher Homologie in den bcl-2 und bcl-xL mRNAs (10). Die Zielsequenz von bcl-2 (605 - 624) ist zu 100% komplementär zu den 20 Basen des 4625 Antisense Nukleotids. Die bcl-xL 
mRNA Zielregion (687 - 706) enthält drei &#8220;Mismatch&#8221; Basen zu 4625. Um die dadurch verminderte Affinität 4625 gegenüber bcl-xL zu erhöhen, wurden beide Oligonukleotide an den 3&#8217; und 5&#8217; Enden chemisch modifiziert. Die unterstrichenen Nukleotide enthalten an der 2&#8217;-Position der Ribose einen MOE Rest. 4625 und 4626 sind vollständige Phosphorothioate. Beide Oligonukleotide wurden mit einem Oligopilot II (Amersham Pharmacia Biotech, Uppsala, Schweden) synthetisiert und durch HPLC aufgereinigt (53). Sie wurden bei -20 °C entweder als Pulver oder in destilliertem Wasser gelöst aufbewahrt.</p>
				</subsection>
				<subsection id="N1074A" label="2.3.3">
					<head>4D5MOC-B-ETA</head>
					<p>4D5MOC-B-ETA wurde als ein rekombinantes Protein in E.coli (SB536) kloniert und die Bakterien in 2dYT Medium (1% Glukose, 30 mg/mL Ampicillin) kultiviert. Die Expression von 4D5MOC-B wurde durch Zugabe von 1mM Isopropyl-D-Galktopyranoside (IPTG, Böhringer Ingelheim) induziert.</p>
					<p>Das Bakterienpellet wurde danach in einer French Pressure Zellpresse (SLS Instruments Inc. Urbana Illinois, USA) lysiert. Das Lysat wurde zentrifugiert und gefiltert (0.22 mm), um es zunächst von groben Verunreinigungen zu befreien. Zur endgültigen Purifikation des Proteins wurden die verschiedenen Fraktionen zuerst durch eine Ni2+ IDA-Säule und dann mit Anionen-Austausch Chromatographie aufgetrennt. Die Reinheit wurde anschliessend im Western Blot überprüft und die Konzentration bei 280 nM spektrometrisch gemessen.</p>
