         <head>Zielsetzung der Arbeit</head>
         <p>
            <citenumber id="N1087A" start="9"/>Das Studium der umfangreichen Literaturquellen, die sich mit der Rasterkraftmikroskopie und verwandten Techniken befassen, lässt sehr schnell erkennen, dass die theoretischen Möglichkeiten der Rasterkraftmikroskopie insbesondere im biologisch- medizinischen Bereich einerseits völlig neue Unter-suchungsansätze- und Philosophien zulassen, andererseits jedoch mit erheblichen präparativen und abbildungstechnischen Problemen behaftet sind. Relativ schnell erreicht man nach Erlernen der rasterkraftmikroskopischen Basiskenntnisse, optimaler Installation des Mikroskops und nach Anwendung einfacher bildverarbeitender Filtertechniken beeindruckende Abbildungen kristalliner Strukturen [z.B. Glimmer = Gruppe gesteinsbildender Silikatminerale (Alumosilikate) mit der allgemeinen chemischen Zusammensetzung M'M3'''((OH)2/AlSi3O10) oder M'M2'''((OH)2/AlSi3O10))] im atomaren Auflösungsbereich. Schnell werden Abbildungsprocedere und Probenpräparation solcher Objekte zur Routine und es gelingen hochauflösende, reproduzierbare Abbildungen dieser Objektoberflächen. Die Ernüchterung nach diesen &#8220;Initialerfolgen&#8221; stellt sich spätestens dann ein, wenn man versucht, morphologische Studien an biologischen Objekten (Zellen, Membranproteine) mit rasterkraftmikroskopischen Methoden durchzuführen. Die Etablierung der objektspezifischen Präparationstechniken und vor allem die Optimierung der vielfältigen AFM-Abbildungsmöglichkeiten gelingt häufig nur nach genauer Analyse der biophysikalischen Eigenschaften der Untersuchungsobjekte. Verallgemeinernd lässt sich formulieren, dass die Erfahrungen, welche man bei der rasterkraftmikroskopischen Abbildung fester, kristalliner Strukturen gewonnen hat, auf weiche, zumeist fluide biologische Objektoberflächen nur begrenzt, um nicht zu sagen kaum anwendbar sind. So ist die Alteration kristalliner Untersuchungsobjekte durch den rasterkraftmikroskopischen Abbildungsprozess vernachlässigbar und damit durch die Auswahl geeigneter Messspitzen leicht zu beherrschen. Im Gegensatz dazu sind biologische Proben während des Abbildungsprozesses höchst vulnerabel. Außerdem ergeben sich Probleme dadurch, dass vitale biologische Systeme (z.B. Zellmembranen und darin eingebettete Struktur- und Funktionsproteine) durch zelldynamische Prozesse charakterisiert sind und demzufolge diese molekularen Strukturen in Abhängigkeit vom Funktionszustand einer ständigen Veränderung unterliegen. Um vitale biologische Strukturen der rasterkraftmikroskopischen Untersuchung zuzuführen, müssen diese auf dem Objektträger immobilisiert werden ohne dabei den Funktionszustand wesentlich zu beeinflussen. Die Qualität der rasterkraftmikroskopischen Abbildung ist also immer ein Produkt aus experimentorientierter optimaler Probenpräparation und der richtigen Auswahl der rasterkraftmikroskopischen Abbildungsparameter. Langfristig erscheint die simultane Visualisierung dieser zelldynamischen Prozesse in Verbindung mit hochaufgelösten morphologischen Strukturanalysen mittels rasterkraft-mikroskopischer Techniken vorstellbar. Essentielle Voraussetzung dafür ist, dass entsprechende Membranoberflächen dieser Zellen der AFM-Abbildung zugänglich gemacht werden.<link id="N1087Dlink"/><link ref="N1087D"><sup class="footnotelabel">67</sup></link>

            <link id="N1089Dlink"/><link ref="N1089D"><sup class="footnotelabel">68</sup></link> Ein Ziel dieser Arbeit soll darin bestehen, kardiovaskulär relevante Zellen in vivo rasterkraftmikroskopisch abzubilden. Die zu untersuchenden Zellen stammen aus etablierten immortalen Kulturzelllinien und aus Primärzellkulturen. Außerdem sollen aortale Endothelzellen im Gewebsverband untersucht werden.<link id="N108BAlink"/><link ref="N108BA"><sup class="footnotelabel">69</sup></link> Neben Endothelzellen unterschiedlicher Herkunft sollen Kardiomyozyten, Fibrozyten und fibroblastenähnliche Zellen (FLC) in vivo rasterkraftmikroskopisch abgebildet werden. Hierbei steht die präparatorische und technisch aufwendige Etablierung der &#8222;Atomic Force Microscopy&#8220; an lebenden, kardiovaskulär relevanten Zellen primär im Mittelpunkt. Die Literatur bezüglich der Rasterkraftmikroskopie an Zellen und zellulären Strukturen verdeutlicht, dass der Etablierungsprozess der Methodik zumeist mit der Abbildung luftgetrockneter Zellen unter Raumbedingungen<link id="N108D7link"/><link ref="N108D7"><sup class="footnotelabel">70</sup></link>

            <link id="N10951link"/><link ref="N10951"><sup class="footnotelabel">74</sup></link> Cryofixation<link id="N10968link"/><link ref="N10968"><sup class="footnotelabel">75</sup></link>), des Abbildungsmodus (Contact-, Non-Contact-, Tapping-Mode) und der Abbildungsbedingungen (Raumbedingungen, zellphysiologische Umgebung) schließlich die Abbildung vitaler Zellen (z.B. kultivierte Zellen aus Pleurapunktat eines Patienten mit metastasierendem Bronchialkarzinom<link id="N10985link"/><link ref="N10985"><sup class="footnotelabel">76</sup></link>) ermöglicht wird. An stabilen oder künstlich stabilisierten Strukturen der Zellen sollen erste orientierende Messungen der bioelastischen Eigenschaften (Kraft-Abstands-Kurven) durchgeführt werden.<link id="N109A2link"/><link ref="N109A2"><sup class="footnotelabel">77</sup></link> Andere hochauflösende strukturanalytische Verfahren (z.B. TEM) erfordern aufwendige Präparationen der zu untersuchenden Objekte und sollen, falls im Einzelfall technisch und apparativ möglich, als Referenzuntersuchungen herangezogen werden. </p>
         <p>Neben der rasterkraftmikroskopischen Darstellung der Oberflächen verschiedener kardiovaskulärer Zellen besteht ein weiteres Ziel dieser Arbeit darin, speziell präparierte Einzelproteine der AFM-Untersuchung zu unterziehen. Ein strukturanalytisch durch Elektronenmikroskopie und Röntgendiffraktionsanalyse sehr gut beschriebenes Funktionsprotein, welches bei der non-lysosomalen, intrazellulären Proteolyse eine entscheidende Rolle spielt, ist das Proteasom. Die 20S-Untereinheit dieses komplexen Proteins soll mittels Rasterkraftmikroskopie abgebildet und vermessen werden. Auf diesem Weg gewonnene strukturanalytische Daten werden mit den bekannten strukturellen Abmessungen des Proteins verglichen und mögliche Abweichungen sollen methodisch diskutiert werden. </p>
         <p>Die Tatsache, dass AFM-Technologie real 3-dimensionale, quasi atomare Auflösung an festen Materialen ohne weiteres ermöglicht, eröffnet ein weiteres, aus kardiologischer Sicht interessantes Anwendungsgebiet. Interventionelle Materialien, die in der täglichen kardiologischen Praxis Anwendung finden sind hinsichtlich ihrer Ultrastruktur und der dadurch beeinflussbaren Gewebskompatibilität häufig nur im Grosstiermodell getestet. Als hochsensitive Methode im Nahfeld von Objektoberflächen bietet die Rasterkraftmikroskopie die Möglichkeit, interventionelle Materialien bezüglich ihrer Oberflächenbeschaffenheit sehr detailliert qualitativ und quantitativ zu beschreiben. Koronare Stents sind heutzutage in der Therapie der koronaren Herzkrankheit nicht mehr entbehrlich. Neben strömungsmechanischen Einflüssen, der Ladungsverteilung<link id="N109C5link"/><link ref="N109C5"><sup class="footnotelabel">78</sup></link> innerhalb der Stentlegierungen (Fe316L-Stahl) und rein hämostaseologischen Kriterien erscheint möglich, dass die zelluläre Adhäsion von Thrombozyten an den Oberflächen der koronarinterventionellen Materialien auch durch die luminale Oberflächentopografie dieser Stents beeinflussbar ist. Die AFM-Untersuchung erlaubt, unterschiedliche Stents hinsichtlich ihrer Oberflächenrauhigkeit sehr präzise zu untersuchen.<link id="N109DFlink"/><link ref="N109DF"><sup class="footnotelabel">79</sup></link> Im Rahmen dieser Arbeit werden diese Analysen an einem klinisch zur Anwendung kommendem Stenttypen (Palmaz-Schatz-Stent) exemplarisch durchgeführt. Es soll untersucht werden, inwieweit das Dilatationsprocedere Einfluss auf die Stentmorphologie hat. Das bedeutet, dass Stents sowohl im nativen Zustand, als auch im dilatierten Zustand untersucht werden. Außerdem sollen im Anschluss an diese Untersuchungen die einzelnen, präparierten Stents mit humanen Thrombozytenkonzentraten inkubiert werden, um zunächst rein morphologisch die Zell-Stentoberflächenkontakte und mögliche, Stentoberflächen- induzierte Veränderungen der Thrombozyten zu beschreiben. Vorarbeiten auf diesem Gebiet sind in der Arbeit von Siedlecki und Marchant beschrieben, die Biomaterial-Zell-Interaktionen auf molekularer Ebene mittels rasterkraftmikroskopischer Techniken untersuchten.<link id="N109F9link"/><link ref="N109F9"><sup class="footnotelabel">80</sup></link> Sollte diese beschriebene Präparation nach entsprechender Zellinkubation rasterkraft-mikroskopisch unter physiologischen Bedingungen darstellbar sein, ergibt sich langfristig die Möglichkeit beispielsweise die thrombozytäre Adhäsionsstärke mit einer modifizierten AFM-Methode, die im angloamerikanischen Sprachraum &#8220;Cell adhesion force microscopy&#8221;<link id="N10A16link"/><link ref="N10A16"><sup class="footnotelabel">81</sup></link> genannt wird, zu untersuchen.</p>
