         <strong>Rezepturen</strong>
         </p>
         <p id="N14A2C">
            <u>Bleizitrat </u>
         </p>
         <p id="N14A32">1g Bleizitrat (Pb<sub>3</sub>(C<sub>6</sub>H<sub>5</sub>O<sub>7</sub>) x 3 H<sub>2</sub>O) wird in 50 ml Aqua dest. gelöst, wobei eine milchige Lösung entsteht. Dieser 2 %igen Bleizitratlösung wird tropfenweise 10 N NaOH-Lösung zugesetzt, bis die Lösung aufklart. </p>
         <p id="N14A44">
            <u>EPON-Gemisch (Kunstharz)</u>
         </p>
         <p id="N14A4A">Benötigte Substanzen:</p>
         <p id="N14A4D">Epon812 (Epichlorohydrin; Epoxyharz)</p>
         <p id="N14A50">MNA (Methylnadic-anhydrid; Härter)</p>
         <p id="N14A53">DDSA  (Dodecenylbernsteinsäureanhydrid; Härter)</p>
         <p id="N14A56">DMP-30 (2,4,6-Tris-(dimethylaminomethyl)-phenol; Beschleuniger)</p>
         <p id="N14A59">17,28 g Epon, 7,92 g MNA und 3,96 g DDSA werden zusammen in einen Plastikbecher gegeben und mit einem Magnetrührer gemischt. Direkt vor Beginn der Einbettung werden  0,6 g DMP-30 hinzufügt und unter Erhitzen wird das Gemisch gerührt bis alles gelöst ist.</p>
         <p id="N14A5C">
            <u>Equithesin</u>
         </p>
         <p id="N14A62">Zur Herstellung von Equithesin werden zunächst 1,15 g Pentobarbital in 12 ml Ethanol  (96 %ig) gelöst. Dann werden 54 ml Propandiol zugegeben, wodurch eine milchige Lösung entsteht (--&gt; Lösung A). Des Weiteren werden 2,5 g Magnesiumsulfat in 50 ml Aqua dest. gelöst (--&gt; Lösung B). Lösung A und B werden nun vermischt, und darin werden 5 g Chloralhydrat gelöst. Die Lösung wird nun durch einen Sterilisationsfilter filtriert und in Portionen zu 20 ml bei 4 ° C gelagert. </p>
         <p id="N14A65">Die Dosierung zur Betäubung des Versuchstieres beträgt 0,0042 ml Equithesin pro 1 g  Körpergewicht.</p>
         <p id="N14A68">
            <u>Farbstoffaliquots</u>
         </p>
         <p id="N14A6E">250 mg Rhodamin-Dextran-Amin (RDA) werden in 1 ml Aqua dest. gelöst. Die Lösung wird in Aliquots zu je 10 µl aufgeteilt. Diese Aliquots werden jeweils mit 10 µl Mineralöl überschichtet, um Sublimieren des Aqua dest. zu verhindern. Zum Aufbewahren werden sie bei &#8211;18 °C tiefgefroren.</p>
         <p id="N14A71">
            <u>Gelatine beschichtete Objektträger</u>
         </p>
         <p id="N14A77">2,5 g Gelatine werden in 500 ml Aqua dest. unter Erwärmen auf  60<sup>0 </sup>C gelöst. Danach werden 0,25 g Chromalaun (Kalium-Chrom-III-Sulfat; Chromium potassiumsulfate) zugesetzt. Die abgekühlte Gelatinelösung wird filtriert.  Die gereinigten Objektträger werden einseitig mit der Gelatinelösung bestrichen. Die bestrichenen Objektträger werden  etwa 30 Minuten im Wärmeschrank bei 40<sup>0 </sup>C getrocknet und ein zweites Mal mit Gelatine bestrichen und getrocknet.</p>
         <p id="N14A80">
            <u>Glutardialdehyd / Paraformaldehyd-Fixativ</u>
         </p>
         <p id="N14A86">9 g Paraformaldehyd werden mit 114 ml Aqua dest. unter ständigem Rühren auf 65 °C erhitzt und gelöst (evtl. ein paar Tropfen 1 N NaOH zufügen, damit die Lösung aufklart). Die auf Raumtemperatur abgekühlte Lösung wird filtriert, dann werden 150 ml 0,2 M Natriumcacodylatpuffer hinzugegeben. Die Lösung wird bei 4 °C aufbewahrt. Kurz vor der Perfusion werden 36 ml einer 30 %igen Glutaraldehydlösung hinzugefügt. </p>
         <p id="N14A89">Zur besseren Membrankonservierung wird der Fixierlösung noch CaCl<sub>2</sub> zugesetzt (Karnovsky 1965). Hierfür werden 2,94 g CaCl<sub>2</sub> in 100 ml Aqua dest. gelöst, davon wird 1 ml der Fixierlösung zugefügt.</p>
         <p id="N14A92">
            <u>Methylenblau-Toluidinblau-Färbelösung</u>
         </p>
         <p id="N14A98">1 Spatelspitze Methylenblau und 1 Spatelspitze Toluidinblau werden in 0,1 M Natriumcacodylatpuffer unter Rühren gelöst. Danach wird die Lösung abfiltriert.</p>
         <p id="N14A9B">
            <u>Natriumcacodylatpuffer</u>
         </p>
         <p id="N14AA1">Für 0,2 M Natriumcacodylatpuffer werden 4,28 g Dimethylarsensäure-Natriumsalz-Trihydrat ( C<sub>2</sub>H<sub>6</sub>AsNaO<sub>2</sub> x 3H<sub>2</sub>O) in 100 ml Aqua dest. gelöst. </p>
         <p id="N14AB0">Für 0,1 M Natriumcacodylatpuffer wird  die 0,2 M Lösung 1:1 mit Aqua dest. verdünnt.</p>
         <p id="N14AB3">
            <u>Nissl-Färbung </u>
         </p>
         <p id="N14AB9">Zunächst wird Acetatpuffer hergestellt. Hierfür werden 16,4 g / l Natriumacetat in Aqua dest. angesetzt und mit 99 %iger Essigsäure auf einen pH-Wert von 6,0 eingestellt. Für die Färbelösung  wird 1 g Kresylviolett in 200 ml Natriumacetatpuffer (pH 3,8 &#8211; 4,0) gelöst.</p>
         <p id="N14ABC">Die Schnitte werden in einer absteigenden Alkoholreihe aus 96 % Ethanol, 70 % Ethanol für je zwei Minuten inkubiert, dann kurz in Aqua dest. gewässert und dann für fünf bis zehn Minuten in der 0,5 %igen Kresylviolett-Acetat-Lösung gefärbt. </p>
         <p id="N14ABF">Zum Ausdifferenzieren der Färbung werden die Schnitte in Acetatpuffer (pH 6,0) gespült und in einer aufsteigenden Alkoholreihe entwässert: 70 % Ethanol, 90 % Ethanol, 96 % Ethanol (jeweils ca. fünf Minuten) und reinem Isopropanol (nur kurz eintauchen). Anschließend werden die Schnitte zweimal für ein bis fünf Minuten in Xylol inkubiert und dann mit Merckoglas eingedeckelt.</p>
         <p id="N14AC2">
            <u>O</u>
            <u>smiumtetroxidlösung (1 %ig)</u>
         </p>
         <p id="N14ACB">Es werden 2 ml einer wässrigen 4 %igen Osmiumtetroxidlösung (OsO<sub>4</sub> ) mit 2 ml 0,2 M Natriumcacodylatpuffer verdünnt. Diese Lösung  wird mit 0,1 M Natriumcacodylatpuffer auf 8 ml aufgefüllt.</p>
         <p id="N14AD1">
            <u>4 % Paraformaldehyd in Phosphatpuffer</u>
         </p>
         <p id="N14AD7">12 g Paraformaldehyd (PFA) werden in 300 ml 0,1 M Phosphatpuffer (PB) bei 60 &#8211; 70 °C gelöst. Wenn die Lösung auf Raumtemperatur abgekühlt ist, wird sie filtiert.</p>
         <p id="N14ADA">
            <u>Phosphatpuffer (0,1 M; pH 7,4)</u>
         </p>
         <p id="N14AE0">14,25 g (NaH<sub>2</sub>PO<sub>4</sub> * H<sub>2</sub>O) und 2,6 g (Na<sub>2</sub>HPO<sub>4</sub> * 2H<sub>2</sub>O) werden in 950 ml Aqua dest. gelöst. Der pH-Wert wird mit NaOH auf 7,4 eingestellt. Danach wird mit Aqua dest. auf 1000 ml aufgefüllt.</p>
         <p id="N14AF5">
            <u>Physiologische Kochsalzlösung (0,9 %)</u>
         </p>
         <p id="N14AFB">9,0 g NaCl werden in 900 ml Aqua dest. gelöst. Der pH-Wert wird mit NaOH auf 7,4 eingestellt. Danach wird die Lösung mit Aqua dest. auf 1000 ml aufgefüllt.</p>
         <p id="N14AFE">
            <u>Saccharose-Lösungen</u>
         </p>
         <p id="N14B04">Zur Herstellung der 15 %igen Saccharoselösung werden 15 g Saccharose in 100 ml Paraformaldehyd (4 %ig in PB) gelöst. Für die 30 %ige Saccharoselösung werden dementsprechend 30 g Saccharose in 100 ml Paraformaldehyd (4 %ig in PB) gelöst.</p>
         <p id="N14B07">
            <u>Uranylacetat</u>
         </p>
         <p id="N14B0D">1g Uranylacetat wird in 50 ml  Methanol (70 %ig) gelöst. Die Lösung wird lichtgeschützt im Abzug aufbewahrt (falls die Lösung trübe geworden ist, muss sie verworfen werden). Vor Gebrauch wird jeweils eine kleine Menge abfiltriert.</p>
