This dissertation from the field of Reflexive Bibliometrics, investigates the constitutive effects of utilizing quantitative metrics in research evaluation on knowledge production and research quality in astronomy. Embedded in the Rational Choice Framework to analyze the structural conditions within academic astronomy, a qualitative analysis reveals an "Evaluation Gap" between what metrics measure and researchers' perceptions of research quality. Furthermore, the analysis unveils a balancing act where astronomers navigate compromises between publication pressures and research integrity. Subsequent quantitative analyses, incorporating Organizational Culture Theories and Self-Determination Theory, demonstrate that controlled forms of motivation exacerbate publication pressure and scientific misconduct, while autonomous motivations yield contrasting effects. Lastly, the study outlines pathways towards transforming research culture towards greater diversity and participation, including the adoption of open knowledge management infrastructures and continuous, reflexive evaluation processes.

Weitere Anwendungsmöglichkeiten werden ausführlich diskutiert.; The first part of this thesis investigates the temporal correlation of photons, generated in a spontaneous parametric down-conversion process inside of a nonlinear crystal, which is placed in a resonator to enhance specific emission lines. However, the cavity influences the temporal correlation of the photons, which is crucial for most applications. This thesis derives a complete theory to describe the temporal correlations of signal-idler, signal-signal and signal-signal-idler photons using the spectral properties of the photon source. The derived theoretical description precisely predicts the experimental measurements, which were performed to verify the theory.
In the second part the exchange phase of photon is measured directly for the first time. Directly, this can only be verified experimentally by interference between the two-photon state and its permuted form. Here a new interferometer technique is introduced to directly determine the photon exchange phase. The experimental results provide evidence of the bosonic nature of photons and state a lower bound for a directly measured exchange phase of photons.
The last part deals with frequency conversion in nonlinear materials. Depending on the wavelengths involved, the conversion processes introduce noise at the target wavelength, which is critical at the single photon level. Then multiple conversion steps are required for a low noise frequency conversion. We present an approach to realize multiple conversion steps with a single nonlinear crystal by applying different local temperatures to that nonlinear crystal. The feasibility of that approach is confirmed experimentally and further possible applications are considered.

The fast detection of explosives is essential to prevent terror attacks. Besides explosives, the trafficking of illegal drugs is a major problem, which must be addressed to reduce criminal organizations’ revenue streams. As low amounts of these substances may indicate trafficking or a bomb threat, sensitive, selective and fast detection methods are required. Therefore, a biosensor was developed in this work, which relies on an immunoassay.
To ensure sensitive and selective performance the used antibodies were characterized in depth. To obtain affinity columns, glass columns were manufactured and used. As column coating affinity conjugates were synthesized. The affinity columns showed efficient antibody trapping and were long-term stable. As a detector an epi-fluorescence microscope was built to monitor the sample. The sensor features consumer-grade parts rendering it easily accessible. For the used fluorophore a limit of detection (LOD) of 2 ppt with a linear range up to 1000 pM was achieved. Due to the use of a syringe pump and an injection valve a continuous fluidic for the biosensor was obtained. The used microfluidic parts resulted in detection times of only 1.5 min.
For TNT a LOD of 130 pM was achieved. Additionally, the biosensor showed no cross-reactivity with common high explosives. For cocaine a LOD of 15 pM in solution was achieved and a surface wipe sampling method was demonstrated, which was able to detect 300 pg cocaine on surfaces within three minutes reliably.
This biosensor belongs to the fastest and most sensitive cocaine and TNT detectors with sensitivity on par or better than liquid chromatography coupled with mass spectroscopy. Furthermore, the biosensor is affordable, very modular analyte-wise and requires no solvents or microplastic carriers. Therefore, it is a fascinating approach for future high-sensitivity monitoring applications.

Direct searches aim to directly observe phenomena, while indirect searches seek discrepancies between theoretical predictions and experimental results. With the discovery of the Higgs boson, the standard model of particle physics was completed, shifting the focus towards indirect searches due to the lack of compelling evidence for new physics at current energy scales. These searches necessitate highly precise theoretical predictions, particularly for non-perturbative hadronic observables, which are calculated using lattice QCD simulations. The need for sub-percent precision has highlighted the importance of accounting for radiative and isospin-breaking corrections, leading to the simulation of fully dynamical QCD+QED.
This thesis addresses the challenges of incorporating QED into lattice QCD, focusing on an approach that maintains gauge invariance, locality, and translational invariance using QED with C-parity boundary conditions (QED$_C$). It presents a comprehensive technical analysis of the first large-scale QCD+QED$_C$ simulations, detailing eight fully dynamical gauge field ensembles with various renormalized electric coupling values ($\alpha_\mathrm{R} \in \{0,1/137,0.04\}$), consistent pion mass ($m_\pi \approx 400$ MeV), and lattice spacing ($a\approx 0.05$ fm). The thesis examines the stability of the simulation algorithm, finite volume effects, and the behavior of different hadron masses.
Furthermore, it elaborates on the tuning of input parameters for lattice simulations to replicate real-world physics accurately, focusing on the hadronic renormalization scheme used to fix bare quark masses. It introduces an optimized strategy for tuning QCD+QED parameters via mass reweighting, adapted for simulations using the RHMC algorithm, highlighting its development, implementation, and testing.

Diese Systeme haben das Potenzial, Phänomene des Standardmodells in einem Niedrigenergieszenario nachzubilden.; Conformal field theory finds applications across diverse fields, from statistical systems at criticality to quantum gravity through the AdS/CFT correspondence. These theories are subject to strong constraints, enabling a systematic non-perturbative analysis. Conformal defects provide a controlled means of breaking the symmetry, introducing new physical phenomena while preserving crucial benefits of the underlying conformal symmetry. This thesis investigates conformal line defects in both the weak- and strong-coupling
regimes. Two distinct classes of models are studied. First, we focus on the supersymmetric Wilson line in N = 4 Super Yang–Mills, which serves as an ideal testing ground for the development of innovative techniques such as the analytic conformal bootstrap. The second class consists of magnetic lines in Yukawa models, which have fascinating applications in 3d condensed-matter systems. These systems have the potential to emulate phenomena observed in the Standard Model in a low-energy setting.

Im Gesamtergebnis liefert diese Dissertation validierte Ansätze zur Erstellung und Einbindung großskaliger Flächen- und Höhendaten städtischer Vegetation, die neue Perspektiven für stadtökologische Fragestellungen bieten.; Urban trees and shrubs have a variety of positive effects on humans and nature, such as cooling overheated inner cities. Detailed information on the location, height, and canopy structure of urban woody plants is indispensable for locating and quantifying such effects, but they are only poorly available on large scales. The globally available digital elevation model (DEM) of the TanDEM-X mission seems suitable for deriving the height and structure of woody areas in cities. This work incorporates TanDEM-X’s DEM with additional remote sensing data and transferable image processing techniques to create urban tree canopy height models (CHM) of woody vegetation and investigate their applicability. The overall goal of this thesis is to provide approaches for creating and applying large-scale, multi-dimensional information on urban wooded areas in a holistic framework. In a first step, CHMs are derived from a TanDEM-X DEM and validated using an additional terrain model for different urban biotopes in Berlin, Germany. Next, first a terrain model and then a crown model are derived in a novel approach, to eliminate the need for an additional terrain model. The accuracies of both steps vary depending on the urban context, but exceed results based on global elevation data with coarser resolution. Then, to generate ideas for future applications of a CHM in urban research, a Berlin CHM is presented to a scientific audience in a written survey. By conducting an urban CHM for the Iranian city of Yazd and integrating it into an urban ecological study, the methodological transferability of the model is tested and the applicability is demonstrated. As an overall result, this thesis provides approaches for the processing and specific application of large-scale area and elevation data on urban vegetation, offering a variety of new perspectives on urban ecological issues.

Darüber hinaus werden Preisdeterminanten des NFT-Marktes analysiert.; This thesis provides models and methodologies developed on robust statistics and their applications in various domains. This dissertation is a historiographical contribution on a sui generis international scholarship program in the form of educational development aid that was implemented between 1977 and 2012 on the second largest island of the Cuban Caribbean archipelago, the Isla de la Juventud. Based on archival sources and reports of participants, Cuba’s motivation for organizing this educational aid is discussed. It is shown that Cuba structured its educational assistance according to the needs and interests of the countries and people involved. It is evidenced that, in addition to the offer of specialties, the program focused on the massive granting of scholarships for lower secondary education and that this measure represented the decisive contribution of the program in view of the low schooling rates in the Third World. Taking into account the main debates on development aid in education, the paper draws on studies of South-South cooperation and the history of official international scholarships to argue that Cuba was guided by the principle of solidarity both in its initial motivation and in the form of the relationships established in the implementation of the program, as well as in the final results of the program. This study concludes that the Cuban development aid policy in the field of education managed to circumvent the “national depersonalization” of the beneficiaries. In other words, participants in the Isla de la Juventud program returned to their countries of origin with a sense of belonging to their nation, taking with them the skills acquired in the ʽopen curriculaʼ and the attitudes conveyed in the ʽnon-written curriculaʼ, which are relevant in different work contexts.

This dissertation examines the life and work of the actress Anna May Wong (1905-1961) through the lens of discourse on race and gender in the USA, Germany, and England. Vice versa: it interrogates the discourse of race and gender by engaging the figure of Wong and her work both on screen and off. Wong’s decades-long career is extraordinary in the sense that, as a Chinese American, she achieved success in the American and European film industry in spite of gendered and racially-based resistance, an experience that her films explore in ambivalent ways. Wong serves as a prism in this dissertation to explore the ways in which racial and gendered boundaries were drawn during the span of her career, but with a specific focus on the films that she made between 1922-1937, which were shot in the United States and Europe. An interrogation of the manner in which Wong appears in these films, as well as the films’ reception(s), reveals a web of complex interweavings, intersections, and ruptures along sexual, gendered, and racialized lines. During the period on which this dissertation focuses, geopolitical power relations between Europe, China, and the United States shifted significantly with dramatic effects on the construction of difference, which the following project explores. The dissertation interrogates, therefore, the relationship between her life and the global context of her work. Her micro-history serves as a fulcrum for examining the larger history of bordering, appropriation, and identity negotiation(s) between Europe, the United States, and China. The investigation of Anna May Wong as a unique phenomenon reveals, on the one hand, a discursive and material history of raced and gendered constructions of difference. On the other hand, her work consistently engages a suspenseful type of excess, which demonstrates the ways in which the “other” is always already embedded in the “self”.

Acquiring a second language (L2) often presents challenges in assimilating the target language's phonological structure, influenced by the native language's (L1) phonology. The phenomenon of phonological interference, wherein the phonological rules of the L1 impact the acquisition of the L2, has been a subject of interest for researchers in the field of second language acquisition. This master's thesis investigates phonological interference between Russian and German, with a particular emphasis on the mechanisms of rule application and ordering. It aims to elucidate the role of rule ordering in Russian to German phonological interference, taking into account rules related to voice alternations, such as final devoicing and regressive devoicing, while also addressing rules unique to the phonology of Russian as the L1. Additionally, the investigation aims to solidify and extend the findings of Rubach (1984) on the role of rule allocation to Lexical Phonology components in phonological interference, by examining cases of Russian to German transfer found in the literature. Expected outcomes from this master's thesis include a deep understanding of Russian phonological rule allocation within the framework of Lexical Phonology. By exploring rule ordering and application in both L1 and L2 phonologies, this study aims to contribute to the development of a unified theory of phonological interference in second language acquisition.

The impact of high-order nonlinear magnetic field errors on the performance of the Large Hadron Collider (LHC) and its planned High-Luminosity upgrade, the HL-LHC, has been extensively studied. Particularly, the presence of such errors in the Insertion Regions (IR) has shown significant repercussions due to the high beta-functions and feed-down to lower orders caused by crossing schemes. This thesis aims to explore different methods for effectively addressing these high-order errors, with the ultimate goal of identifying and correcting them to optimize beam optics and enhance machine performance.
Simulation studies are employed, using a novel and flexible correction algorithm developed during the course of this PhD research. Various strategies are investigated to improve corrections by targeting Resonance Driving Terms (RDTs) associated with diverse error sources.
Special attention is devoted to decapole and dodecapole errors,
which have demonstrated detrimental effects on amplitude detuning due to feed-down
based on previous measurements in the LHC. The anticipated increase in optics sensitivity to errors in the IRs of the HL-LHC further underscores the importance of addressing these errors.
Correction options are evaluated, focusing on the utilization of the nonlinear corrector packages to address errors in the new separation and recombination dipoles in the HL-LHC, where increased decapole errors had been expected.
Experimental studies are conducted to validate the findings. Significant efforts are dedicated to mitigating the feed-down effects arising from decapole and dodecapole field errors. To address this challenge, novel corrections involving the operational implementation of dodecapole correctors in the IRs have been introduced for the first time.
The results of these experiments provide valuable insights into the mitigation of high-order errors and contribute to the overall understanding of beam dynamics in advanced particle accelerators.

In the first part, a comparison of sample preparation and instrumental methods for organofluorine determination was used to identify advantages and disadvantages in sum parameter analysis of per- and polyfluoroalkyl substances (PFASs). Therefore, surface water samples from the Spree River in Berlin, Germany and industrial effluent samples were analyzed using two PFAS sum parameters – the extractable (EOF) and adsorbable organically bound fluorine (AOF). Both PFAS sum parameters were analyzed using two fluorine sensitive detection systems – high-resolution-continuum source-graphite furnace molecular absorption spectrometry (HR-CS-GFMAS) and combustion ion chromatography (CIC). HR-CS-GFMAS turned out to be the more sensitive and faster approach accompanied with higher precision compared to CIC.
In the second part, a new method for PFAS sum parameter analysis and EOF determination in soil samples based on HR-CS-GFMAS was developed and optimized. EOF mass fractions were investigated upon multiple extraction and utilization of solid phase extraction (SPE) for inorganic fluorine removal. In a comparison of the optimized method with and without SPE, a highly biased discrimination of organofluorines using SPE was shown.
In the third part, organofluorine mass balances were investigated in aquatic systems using surface water and suspended particulate matter (SPM) samples. SPM samples were analyzed using 4 complementary analytical methods – target and non-target HRMS, direct total oxidizable precursor assay (dTOPA) and EOF. Therefore, 3 organofluorine mass balance approaches were used for SPM sample analysis in spatial and time resolved manner: (I) Identification of oxidizable PFAS precursors, (II) Identification of the proportion of identified and unidentified EOF and (III) PFAS target, PFAS dTOPA, non-target screening and EOF analysis to further unravel the unidentified EOF from approach (II).

The aim of this doctoral work is to provide a comprehensive study and overview on the topic of the heterogeneity of microglia in the normal adult mouse brain from the morphological, electrophysiological and transcriptional perspective with the focus on differences between white and grey matters. In the first Chapter, I discuss the morphological heterogeneity of
microglia in the brain with the focus on two morphologically distinct classes: satellite and parenchymal microglia. We performed an in-depth analysis of multiple brain areas and quantified the number of satellite microglia which is in contact with different neuronal subtypes. We found that satellite microglia numbers are highly correlated with neuronal densities of a certain area, while showing no preferences for any of the neuronal types.
In Chapter two of this work, I study transcriptional heterogeneity of microglia from white and grey matters. For this I am employing Patch-seq, which we newly established in glial cells. This method allows a combination of morphological, electrophysiological and transcriptional profiles of single cells to assess their differences. When comparing Patch-seq dataset to the previously published FACS isolated single cell RNA-seq microglia datasets, we find a subtype of cells which uniquely belongs to FACS sample and is characterized by expression of stress-associated genes. This finding points out to the fact of dissociation-related artifacts in the single cell RNA-seq data which are not present in situ.
In the third chapter, I identified transcripts which are induced during the dissociation of the tissue by employing the SLAM-seq method. This procedure allows to metabolically label newly transcribed mRNA and computationally remove transcripts from the sample. By removing the labeled transcripts from the dataset of cells isolated from the hippocampus via enzymatic dissociation, we observe that an “activated microglia” subtype merges with the general microglia population.

Alle Deformationen ergeben die gleiche Streumatrix, die mit der erwarteten exakten Streumatrix bei trigonometrisch quantendeformierter Symmetrie übereinstimmt.; We study the perturbative quantum theory of various integrable Yang-Baxter deformations of the free superstring on AdS spaces. For this we compute the two-body tree-level scattering matrix on the worldsheet using Feynman diagram methods.
The various deformations are: (1) All distinct Abelian deformations of AdS₅ ⨉ S⁵ allowing light-cone gauge fixing. These are dual to noncommutative super Yang-Mills theory and equivalently described through TsT transformations or twisted boundary conditions.
We compute the bosonic tree-level scattering matrix for the BMN string in uniform light-cone gauge. The scattering matrix is expressed through a Drinfeld twist for most cases;
for some cases it is expressed instead through a shifted momentum dependence.
Lastly, we compare the Bethe equations derived from these results to the equations of the model with twisted boundary conditions and find perfect agreement.
For deformations of the GKP string we are not able to determine a deformed scattering matrix around the null-cusp solution due to actions incompatible with perturbation theory in momentum space. (2) The inhomogeneous or eta deformation of AdS₅ ⨉ S⁵ corresponding to the fermionic Dynkin diagram. We compute the two-body tree-level scattering matrix up to second order in fermions in uniform light-cone gauge.
It satisfies the classical Yang-Baxter equation, factorizes into two blocks and matches the exact scattering matrix for a model with trigonometrically quantum-deformed symmetry.
(3) Inhomogeneous bi-Yang-Baxter deformations of AdS₃ ⨉ S³ ⨉ T⁴ for multiple Dynkin diagrams. We compute the two-body tree-level scattering matrix up to second order in fermions in uniform light-cone gauge. All deformations give the same scattering matrix, which matches the expected exact scattering matrix with trigonometrically quantum-deformed symmetry.

This thesis, based on personal experience as an asylum seeker in Germany, focuses on the socio-economic integration of Black African asylum seekers, i.e. with a residence permission § 55 of the German Asylum Act, as well as tolerated migrants, i.e. with a toleration status § 60a German Residence Act. The thesis sheds light on their participation in German integration courses and their access to work permits across the sixteen German states, which are subject to strict regulations by the Foreigner Registration Office and the Federal Office of Work.
The findings reveal that individuals engage in economic activities to financially sustain themselves within the territory and also to support family members in their home countries.
Methodically, the thesis employs mixed methods, utilizing ethno-sociogeographical tools for data acquisition, processing, and analysis. Between 2016 and June 2021, the thesis conducted twenty individual case studies, three hundred semi-structured questionnaires, two expert interviews with members of German parliaments, six focus group discussions with the target group and German officials from the court, the Foreigner registration office. The economic activities uncovered include drug dealing, newspaper delivery, informal street food selling, hairdressing, trading, and COVID-19 testing. As there is a massive use of informal economic practices (illegal, semi-legal, and criminal) as entry point to the job market and as a response to limited opportunities provided by the German authorities, the thesis concludes with a call to policymakers to urgently reconsider the current socio-economic integration framework for this two categories of migrants in Germany.

We created a multi-omic virtual Drosophila melanogaster stage 6 embryo by simultaneously assessing genome accessibility and transcriptional states in individual cells. Using novoSpaRc, a spatial mapping tool, we accurately reconstructed the spatial origin of cells, yielding a regulatory blueprint reflecting gene expression and enhancer accessibilities. This resource aids in understanding developmental regulatory dynamics.
Examining ATAC-peaks, we observed overlapping chromatin accessibility patterns with the activity of independently testes enhancers, emphasizing accessibility's importance. Non-negative matrix factorization identified archetypes in gene expression and chromatin accessibility. Accessibility archetypes, potentially regulated by transcription factors (TFs), were subjected to motif enrichment analysis for archetype-associated CRMs. An enhancer prediction approach, utilizing a generalized linear model, assigned enhancers to genes based on partial pattern similarity.
In summary our multi-modal virtual embryo serves as a resource and presents for the first time
single-cell chromatin accessibilities for genomic regions reconstructed in space for a whole organism in
a single developmental stage. The results shed light on gene regulatory principles, highlighting the regulatory impact of TFs on chromatin states of enhancers.

Was sind Diffusionsmodelle?
How do they work? Can they improve image classification
algorithms? These are the main questions I will try to answer in this thesis. I will try to label
different images of statistical charts that originate from the open-source library Wikimedia
Commons. In total, this library contains almost 100 million media elements. A loose policy
regarding the creation of categories and the uploading of images has led to a lot of unlabeled or falsely classified images. Another challenge when labeling the images on Wikimedia Commons is that some niche classes are very small.
I will tackle these problems by training image classification algorithms on cleaned data sets of selected categories. I will enhance the training and validation sets with different amounts of artificial images. While there are many exciting results, the most important one is that generative data augmentation seems to work well for major categories but not that well for niche categories, which are similar conceptually.

The small intestinal epithelium is the primary route of infection for many protozoan parasites such as Toxoplasma gondii and Giardia duodenalis. Understanding the mechanisms of infection with such parasites, has been hindered due to the lack of appropriate models mimicking the complexity of the intestinal epithelium. Here, an in vitro platform was established, using intestinal organoids (organoid derived monolayers or ODMs) for comparative studies on parasite-host interactions.
The ODM system was used to model the events during the early phase of a T. gondii infection, focusing on the role of Interferon gamma (IFNγ) and Immunity Related GTPases (Irgs). Irg dependent control of virulent Toxoplasma strains has been shown to differ between the laboratory mouse model and other wild-derived rodent strains. How these responses occur in rodent species that do not belong to the murine family, is yet to be determined. For this reason, comparisons were made with organoids from different laboratory mice strains and the bank vole Myodes glareolus, a non-muridae rodent species assumed to be more resistant to T. gondii.
Based on this thesis, stimulation with IFNγ leads to a trend of reduced replication in M. glareolus ODMs compared to Mus ODMs for both Type I parasite strain RH and for Type II strain Prugniaud, based on quantitative immunofluorescence assays and qPCR. Analysis of the role of Irgs in intestinal T. gondii infections was performed, by transfecting organoids from M. glareolus with a fluorescently labelled Irgb10 protein, showing that Irgb10 decorates T. gondii vacuoles, suggesting Irg-system involvement.
Finally, a co-infection model in murine ODMs was established for T. gondii and/or G. Duodenalis. Here, it could be observed that T. gondii did not influence G. duodenalis induction of barrier breakdown nor did G. duodenalis influence T. gondii replication.

Recent advances in econometric theory and machine learning methods have allowed researchers to adpot and develop new approaches for empirical analyses, some of which will be discussed in this thesis.
Chapter 3 is about predicting merger & acquisition (M&A) events. While there is no doubt that M&A activity in the corporate sector follows wave-like patterns, there is no uniquely accepted definition of such a "merger wave" in a time series context. Count-data time series models are often employed to measure M&A activity and merger waves are then defined as clusters of periods with an unusually high number of M&A deals retrospectively. However, the distribution of deals is usually not normal (Gaussian). More recently, different approaches that take into account the time-varying nature of M&A activity have been proposed, but still require the a-priori selection of parameters. We propose adapating the combination of the Local Parametric Approach and Multiplier Bootstrap to a count data setup in order to identify locally homogeneous intervals in the time series of M&A activity. This eliminates the need for manual parameter selection and allows for the generation of accurate forecasts without any manual input.
Chapter 4 is an empirical study on jumps in high frequency digital asset markets. While attention is a predictor for digital asset prices, and jumps in Bitcoin prices are well-known, we know little about its alternatives. Studying high frequency crypto ticks gives us the unique possibility to confirm that cross market digital asset returns are driven by high frequency jumps clustered around black swan events, resembling volatility and trading volume seasonalities. Regressions show that intra-day jumps significantly influence end of day returns in size and direction. This provides fundamental research for crypto option pricing models and opens up possibilities to evolve econometric theory to better address the specific market microstructure of cryptos.
Chapter 5 discusses the increasing adoption of Digital Assets (DAs), such as Bitcoin (BTC), which raises the need for accurate option pricing models. Yet, existing methodologies fail to cope with the volatile nature of the emerging DAs. Many models have been proposed to address the unorthodox market dynamics and frequent disruptions in the microstructure caused by the non-stationarity, and peculiar statistics, in DA markets. However, they are either prone to the curse of dimensionality, as additional complexity is required to employ traditional theories, or they overfit historical patterns that may never repeat. Instead, we leverage recent advances in market regime (MR) clustering with the Implied Stochastic Volatility Model (ISVM) on a very recent dataset covering BTC options on the popular trading platform Deribit. Time-regime clustering is a temporal clustering method, that clusters the historic evolution of a market into different volatility periods accounting for non-stationarity. ISVM can incorporate investor expectations in each of the sentiment-driven periods by using implied volatility (IV) data. In this paper, we apply this integrated time-regime clustering and ISVM method (termed MR-ISVM) to high-frequency data on BTC options. We demonstrate that MR-ISVM contributes to overcome the burden of complex adaption to jumps in higher order characteristics of option pricing models. This allows us to price the market based on the expectations of its participants in an adaptive fashion and put the procedure to action on a new dataset covering previously unexplored DA dynamics.

Dieser Algorithmus, der zusammen mit der AG Marsico (ICB - Helmholtz Zentrum München) entwickelt wurde, sagt RNA-Bindeproteine voraus, mithilfe von Tripeptiden in intrinsisch ungefalteten Regionen (IDR). 66 RBPs hat TriPepSVM in Salmonella vorausgesagt, wovon drei im Rahmen dieser Arbeit experimentell validiert wurden.; Salmonella enterica is a species of Gram-negative bacteria and the causative agent of enteric diseases, ranging from gastroenteritis to systemic infections, causing hundreds of thousands of deaths every year.
In the last 30 years the basic mechanisms underpinning invasion and intracellular growth have been unravelled: Salmonella produces tens of virulence factors - termed “effectors” - which are secreted by two distinct Type III Secretion Systems (T3SS) for the hijacking of the host cell molecular machinery via protein-protein interactions. Although the biochemical activities of many effectors have been characterised, the functions of some have remained elusive. In addition, post-transcriptional regulation has emerged to prominence in bacteria, with RNA-protein interactions playing a pivotal role in many bacterial functions, including infection.
One of the most frequently used techniques to study Salmonella effectors is the Gentamicin Protection Assay (GPA), a simple method which replicates the infection process in vitro; however, as GPA is subject to high levels of variation and only generates end-point measurements, the method can be inadequate when studying multiple conditions at once or following time-dependent dynamics. InFluence, a high-throughput, fluorescence microscopy-based analysis pipeline, was designed to address these issues. InFluence offered insights into the intracellular niches occupied by Salmonella, and proved to be a useful tool in assessing not only the contribution to the infection process of effectors, but that of host proteins too.
In addition, we contributed to the development of TriPepSVM, a support vector machine-based (SVM) method developed with the Marsico group (Institute of Computational Biology (ICB) - Helmholtz Zentrum Munchen) for predicting RNA-binding proteins (RBPs) using tripeptides that are frequent in Intrinsic Disordered Regions (IDRs). TriPepSVM predicted 66 new RBPs in Salmonella, and three were experimentally validated.

Recent descriptions revealed unique features such as large fangs and a gland in the lower jaw containing volatile hydrocarbons resembling insect pheromones. This dissertation aimed to formalize the description of the gland and ascertain its function. The first chapter delved into to frogs’ biology, unveiling site loyalty, and territorial behaviors, especially in males. Injuries suggested aggressive interactions, particularly around breeding habitats. The frogs use water-filled crevices for egg deposition, hinting at complex mating and habitat interactions. Two main hypotheses emerged regarding gland function: intraspecific communication for coordination during oviposition and fertilization, or interspecific communication to attract prey. The second chapter explored the diet to support the latter hypothesis. Findings showed a generalist diet and significant amounts of leaflets from a specific tree, indicating an unprecedented plant uptake. This refuted the idea of attracting specific prey and supported intraspecific communication. The third chapter focused on the acoustic repertoire, revealing a species-specific advertisement call in males and unexpected squeaking calls in females. Contrary to assumptions, acoustic communication in Saber-toothed frogs was pronounced and not restricted. The fourth chapter examined the gland's anatomy and chemical composition. The gland, present in juveniles and varying in adults, exhibited species-specific compositions varying with sex and reproductive state. In conclusion, Saber-toothed Frogs most likely employ multimodal communication, combining acoustic and chemical signals to coordinate mating and oviposition. The unique gland and its secretion hint at a more widespread use of chemical communication in frogs than previously recognized.

This dissertation examines various aspects related to maternal employment, fertility and the skill development of children in Germany.
In chapter two, we develop an estimation method for the causal effect of the 2007 parental benefits reform in Germany. Therefore, we first estimate the "child penalty'' on employment outcomes, then we use the estimated child penalties before and after reform implementation to assess reform effects. We find that the reform had positive effects on medium-run employment.
Chapter three focuses on the so-called "cash-for-care''-transfers for parents of children aged one to two. I find a significant reduction of employment for migrant mothers if the potential benefit amount is increased. There are positive effects on fertility for the average of the population. However, I do not find significant effects on the skill development of children.
In chapter four, we study the expansions of German all-day schools and their impact on children's outcomes. Our findings reveal evidence of positive impacts on children's achievements. We also show significant positive peer effects from classmates attending all-day programs. However, we do not find significant evidence that these programs significantly contribute to decreasing inequality in the German school system.
Chapter five addresses an important aspect in the administrative labor market data. By developing a correction for misreported part-time employment spells which happened in the years prior 2011. The corrected data have implications for studying wage inequality, but also for studying maternal employment as the part-time share is very high among mothers of young children.
In summary, this dissertation studies the period from birth to primary school. It covers topics such as maternal employment, fertility and skill development. Additionally, it addresses the data problem of misreported part-time spells in the administrative labor market data and presents a correction methodology.

Die Ergebnisse werden durch verschiedene Data-Snooping-Tests überprüft.; This dissertation shows the potential of machine learning methods for managing tail risk in a non-stationary and high-dimensional setting. For this, we compare in a robust manner data-dependent approaches from parametric or non-parametric statistics with data-adaptive methods. As these methods need to be reproducible to ensure trust and transparency, we start by proposing a new platform called Quantinar, which aims to set a new standard for academic publications. In the second chapter, we dive into the core subject of this thesis which compares various parametric, local parametric, and non-parametric methods to create a dynamic trading strategy that protects against tail risk in Bitcoin cryptocurrency. In the third chapter, we propose a new portfolio allocation method, called NMFRB, that deals with high dimensions thanks to a dimension reduction technique, convex Non-negative Matrix Factorization. This technique allows us to find latent interpretable portfolios that are diversified out-of-sample. We show in two universes that the proposed method outperforms other classical machine learning-based methods such as Hierarchical Risk Parity (HRP) concerning risk-adjusted returns. We also test the robustness of our results via Monte Carlo simulation. Finally, the last chapter combines our previous approaches to develop a tail-risk protection strategy for portfolios: we extend the NMFRB to tail-risk measures, we address the non-linear relationships between assets during tail events by developing a specific non-linear latent factor model, finally, we develop a dynamic tail risk protection strategy that deals with the non-stationarity of asset returns using classical econometrics models. We show that our strategy is successful at reducing large drawdowns and outperforms other modern tail-risk protection strategies such as the Value-at-Risk-spread strategy. We verify our findings by performing various data snooping tests.

However, during the fabrication process, several defects are introduced to the sensor surface, resulting in complex surface properties. The large-area silicon sensors for the ATLAS Inner Tracker (ITK) Upgrade show signs of early surface breakdown at high humidity. This results in the sensors experiencing a high leakage current and are not inoperable as particle detectors when biased in a high-humidity environment. To investigate the surface properties and understand the breakdown mechanism, the Top Transient Current Technique (Top TCT) is used to study the charge transport process in silicon sensors. This method utilizes fast laser pulses, focused to a micrometer-sized spot, to illuminate the front side of the sensor, generating electron-hole pairs within the first several micrometers from the surface. These charge carriers then drift under the influence of the applied electric field, inducing transient currents on the electrode. By analyzing these transient signals as a function of the laser-injected position and the applied bias voltage, the distribution of the surface electric field can be qualitatively estimated. In this work, both simulation and measurement using the Top TCT method on a planar diode are performed. The simulation is conducted using the Allpix Squared software, a Monte Carlo simulation framework specially developed for semiconductor detectors. The effective doping concentration, full-depletion voltage, and active thickness of a planar silicon n-in-p diode are measured using the Top TCT method. The transfer function of the read-out electric circuit is extracted through Fourier transform analysis of both the simulated and measured data. A new parameter ”surface reflectivity” is implemented and modified in Allpix Squared to simulate the charge transport at the silicon-silicon dioxide interface, where a field pushing the electrons toward the interface exists. Laser scans between the edge ring and guard ring of the diode for different bias voltages are simulated and measured. By comparing the simulation results to the measurement in aspects of the transient pulses, velocity profile and charge profile, a better understanding of the surface properties of a silicon diode can be obtained, from which the humidity dependence on the surface breakdown may be explored.

However, hydrological quantifications are challenging due to the spatial heterogeneity in environmental drivers of the underlying processes. Therefore in this thesis, a series of common linked approaches or technologies were developed and tested in a mixed-landuse catchment, the Demnitzer Millcreek (DMC, 66 km2). By applying data analysis, distributed modelling, machine-learning, and unmanned-vehicle technologies, we not only unraveled the hydrological processes in this representative catchment, but also from a wider perspective - identified the optimal way to jointly use the existing technologies to reduce the uncertainty in monitoring and modelling these processes at the catchment-scale.
